SAMMEND: Age determination at death of skeletal remains can prove difficult.
The teeth are often well preserved and could present the only means
for an age evaluation. Based on the closureo f skull sutures and
dental attrition, the age of a Stone Age man was judged to be about
60 years when he succumbed. This has now been verified by means of
measuring the extent of roo t dentin transparency. The importance of
teamwork in difficult cases regarding assessment of age in skeletal
remains is pointed out.

SAMMEND: The sequence of the omp1 gene coding for the 40-kDa outer membrane
protein (OMP) of the Gram- oral bacterium. Fusobacterium nucleatum
strain Fev1, has been determinded. Degenerate
oligodeoxyribonucleotide primers were used to prime the amplification
of a 120-mer sequence of the gene. This sequence was successively
used for constructing new primers applied in asymmetrical,
symmetrical, and inverse polymerase chain reaction using as template
genomic DNA, self-ligated DNA fragments, or fragments ligated into
either pGEM-7Zf+ or pACYC184. The codon usage of the gene was
unusual in that A or T was used as the third base in the codon
triplets in all cases, except for those amino acids (aa) which have
only one or two possibl e codon choices. Only 35 of the 61 sense
codons were used. The aa sequence of the protein was deduced; it
consisted of 348 aa (M r 39954) , which is in good agreement with the
40-kDa size estimated from electrophoretic analyses. The mature
protein was preceeded by a 20-aa signal peptide.

SAMMEND: DNA from six strains of Fusobacterium nucleatum was examined for
possible methylation of adenine or cytosine in the restriction site
GATC. F. nucleatum DNA was digested with the restriction enzymes
Sau3Al, Dpnl and Mbol, subjected to electrophoresis in agarose gels
and Southern blotting. The blots were hybridized with a polymerase
chain reaction (PCR) -amplifie d DNA probe. Both radioactive and
non-radioactive labeling methods were used. Some strains of F.
nucleatum were found to be dam methylated or dcm methylated in the
sequence GATC;thea denine residue is methylated in strain ATCC 10953,
while the cytosine residue is methylated in the strains F1 and F3.
Strains F6, ATCC 25586 and Fev1 are probably not methylated in t his
nucleotide sequence.

SAMMEND: A polymerase chain reaction probe with 100% sequence identity to 120
deoxyribonucleotides of Fusobacterium nucleatum Fev1, coding for a
part of the 40-kDa major outer membrane protein, was labeled with the
steroid hapten digoxigenin. The probe was compared with various
degenerate oligonucleotide probes and found to tolerate much more
stringent washing condition s. It was therefore superior in
distinguishing, by means of Southern blots and slot blots,
F.nucleatum from other oral gram-negative bacteria in the periodontal
pocket and from other fusobacterial species and in distinguishing
among different strains of F. nucleatum. F. periodonticum was found
to be more similar to F. nucleatum than the other fusobacterial
species tested.

SAMMEND: Lipopolysaccharides (LPSs) from anaerobic gram-negative bacteria,
including those of low endotoxic activity that are isolated from
Bacteroides, Prevotella, and Porphyromonas are potent inducers of DNA
replication and polyclonal immunoglobulin production in murine B
lymphocytes. The activation in dose-dependent and T cell-independent.
Replication of DNA and production of immunoglobulins were also
stimulated by lipid A and by the LPS heteropolysaccharide that were
isolated by mild acid hydrolysis of the LPSs of Bacteroides fragilis
and Fusobacterium nucleatum. Combinations of LPS, lipid A, and
acid-degraded polysaccharide amplified the blastogenic response.
Antibodies that react with the polysaccharide part of LPSs isolat ed
from members of the Bacteroidaceae are present in healthy human
serum.

SAMMEND: Fluorescent microspheres (FM) were used to visualize and semiquantify
blood flow in the temporomandibular joint (TMJ) during experimental
unilateral traumatic occlusion of the maxillary and mandibular molar
teeth in 30 young rats. At different postoperative observation
periods varying from 1 to 30 days FM were injected systemically, and
the number of FM were counted in serial sections from the TMJ in a
fluorescent microscope. Blood flow was related to the number of FM
found in the fibrous connective tissue and bony condyle of the TMJ. A
percent age increase in blood flow was found at 15 to 20 days on the
experimental side, compared with the contralateral side. Furthermore,
there was an increase in blood flow in both TMJs in the experimental
animals compared with an unoperated control material of 10 animals.
The study thus indicates that a local unilateral occlusal trauma
initiates blood flow respons es not only unilaterally but also in the
TMJ on both sides in rats.

SAMMEND: Sorghum and maize porridge are used as infant weaning foods in many
African countries. Porridges prepared from the cereals have high
viscosity; to reduce viscosity, the cereals are initially sprouted
and made into flour or fermented before use. This paper compares the
effect of these preparatory methods on the proximate composition,
mineral and fatty acid characteristics of the sprouted and/or
fermented sorghum and maize. The preparatory methods had no effect on
the proximate composition. The cereals are poor in calcium, iron and
zinc. Theya re low in w3 fatty acids but rich in the w6 fatty acids.
Germination increased the gross energy of the cereals. Porridges
prepared from these cereals need to be supplemented by othe r foods.

SAMMEND: Sprouted white maize meal was supplemented with 5, 10, 15 and 20%
(w/w) sundried pelagic fish meal. The nutritive value for young rats
fed gruels of these mixtures was determined.The protein, fat, ash,
and energy contents of the dried gruels increased when the amount of
fish in the diet increased. Both supplemented and unsupplemented
samples showed high true di gestibilities (above 90) and there was
no statistical difference between them. NPU0 and NDp-Cal% increased
with increasing amount of fish in the diet. Supplemented maize meals
at t he 10, 15 and 20% levels were better utilized than the
unsupplemented diet (p<0.05). Furthermore, a decreased in-vitro
protein synthetic capacity was shown in animals fed low protein
diets. There was good correlation (r=0.90) between weight gain and
synthetic activity. Supplementation of sprouted white maize meal with
5-10% fish protetin was sufficient to optimi z e the protein
utilization in young rats.

SAMMEND: The small pelagic fish caught in the freshwaters of Africa are mostly
sundried and used in meal preparation. They are not generally used in
weaning foods. With a view to incorporat ing fish into weaning diets,
the nutritional quality of fish samples from the lakes along the Rift
Valley in Africa was assessed. Fish are good sources of protetin,
ash,and fat. Theyare poor in vitamins A and E and their contribution
to the diet will depend on the amount eaten. Fish lipids are good
sources of essential fatty acids. Docosahexaenoic acid and eicosa
pentanoic acid which are important in brain development are present
in substantial amounts. Fish is generally low in thiamine and
pyridoxine but high in niacin. The content of calcium in whole fish
is very high and there are appreciable quantities of zinc and iron.
The amino acid profiles are in good agreement with the FAO
provisional pattern and to the requireme nts of infants. The limiting
amino acid is tryptophan.

SAMMEND: Holes drilled in rats' skull, iliac crest, and tibia were filled with
beeswax or with new, wax-like, bioerodible polyorthoester (Alzamer
(R)). Empty drill-holes served as controls. In addition, beeswax and
polyorthoester were deposited between the left and the right oblique
abdominal muscles, respectively. In muscle, both the beeswax and
polyorthoester elicited a transient foreign body reaction. The
beeswax was not resorbed in bone or muscle, whereas the
polyorthoester was. Bone healing was inhibited in the iliac crest and
the tibiae filled with beeswax, whereas holes filled with
polyorthoester healed readily.
